To measure surface area, use the Freehand Tool from the ImageJ toolbar. Click and drag the line around the area you which to measure. Click on Analyze > Measure (or push Apple-M) to find the surface area of the shape that was drawn. To get rid of the Freehand Tool, click anywhere on the image once.
